TMZ said Murray entered the plea Tuesday during his arraignment.
"Your Honor, I am an innocent man," Murray told Los Angeles County Judge Michael Pastor. "I, therefore, plead not guilty."
Murray's trial is set to begin March 28.
The cardiologist is accused of causing Jackson's death by improperly administering sedatives and anesthesia in an effort to help the singer sleep. Jackson died at age 50 from acute propofol intoxication June 25, 2009.
